Das Humane KL-6 (Krebs Von den Lungen-6) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsquantifizierung von KL-6.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes KL-6 ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, dann mit einem biotinylierten Antikörper, der spezifisch für humanes KL-6 ist. Als Nächstes wird an Meerrettichperoxidase (HRP) konjugiertes Avidin zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die humanes KL-6, biotinylierten Antikörper und enzymkonjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem KL-6 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
