Das Human CLEC12B(C-Type Lectin Domain Family 12, Member B) ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Messung von CLEC12B in der Forschung.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Antikörper vorbeschichtet, der spezifisch für humanes CLEC12B ist. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, danach ein biotin-konjugierter Antikörper, der spezifisch für humanes CLEC12B ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die humanes CLEC12B, biotin-konjugierten Antikörper und enzym-konjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung gestoppt, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem CLEC12B in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
