Das Humane PPP2R4 (Proteinphosphatase 2A-Aktivator, regulatorische Untereinheit 4) ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Forschungsuntersuchung von PPP2R4.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Human PPP2R4-spezifischen Antikörper vorbeschichtet. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für humanes PPP2R4 ist. Als Nächstes wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die humanes PPP2R4, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Human PPP2R4 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
