Das Maus-HbA1c(glykiertes Hämoglobin A1c) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von HbA1c.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus-HbA1c ist. Standards oder Proben werden dann mit einem Biotin-konjugierten Antikörper, der spezifisch für Maus-HbA1c ist, zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt. Als Nächstes wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nachdem die TMB-Substratlösung hinzugefügt wurde, zeigen nur die Vertiefungen, die Maus-HbA1c, biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch die Zugabe von Schwefelsäurelösung beendet und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-HbA1c in den Proben wird dann durch den Vergleich der OD der Proben mit der Standardkurve bestimmt.
