Dieses Assay-Kit basiert auf dem Sandwich-ELISA-Prinzip. Die PVDF-Membran am Boden der Mikro-ELISPOT-Platte wurde mit anti-humanen IL-1β-Antikörpern neu beschichtet. Während des Experiments werden zuerst Zellsuspension und Stimulanzien hinzugefügt. Das von den Zellen während des Inkubationsprozesses sezernierte IL-1β bindet an die beschichteten Antikörper. Anschließend werden die Zellen abgewaschen und nacheinander biotinylierte anti-humane IL-1β-Antikörper und alkalische Phosphatase-konjugiertes Avidin hinzugefügt. Die biotinylierten anti-humanen IL-1β-Antikörper binden spezifisch an das bereits gebundene IL-1β, woraufhin Avidin spezifisch an Biotin bindet und so den Komplex bildet. Nachdem freie Komponenten abgewaschen wurden, wird chromogenes Substrat (BCIP/NBT) hinzugefügt. BCIP unterliegt unter der Katalyse der alkalischen Phosphatase einer Hydrolyse und reagiert mit NBT, wodurch unlösliche Spots mit dunkelblauer bis blau-violetter Farbe entstehen. Zur Analyse des Ergebnisses kann eine professionelle ELISPOT-Plattenlesesoftware verwendet werden, die die Frequenz der humanen IL-1β-sezernierenden Zellen berechnet.
