Technical note
Dieses CLIA-Kit nutzt das Sandwich-CLIA-Prinzip. Die in diesem Kit enthaltene Mikro-CLIA-Platte ist mit einem Antikörper vorbeschichtet, der spezifisch für Maus F8 ist. Proben oder Standards werden in die Vertiefungen der Mikro-CLIA-Platte gegeben und mit dem spezifischen Antikörper kombiniert. Anschließend werden nacheinander ein biotinylierter Nachweisantikörper, der spezifisch für Maus F8 ist, und ein Avidin-Meerrettichperoxidase (HRP)-Konjugat in jede Mikrotiterplattenvertiefung gegeben und inkubiert. Freie Komponenten werden abgewaschen. Die Substratlösung wird jeder Vertiefung hinzugefügt. Nur die Vertiefungen, die Maus F8, den biotinylierten Nachweisantikörper und das Avidin-HRP-Konjugat enthalten, zeigen Lumineszenz. Der Wert der relativen Lichteinheit (RLU) wird mit dem Chemilumineszenz-Immunoassay-Analysator bei 425 nm gemessen. Der RLU-Wert ist positiv mit der Konzentration von Maus F8 korreliert. Sie können die Konzentration von Maus F8 in den Proben berechnen, indem Sie den RLU-Wert der Proben mit der Standardkurve vergleichen.
