Technical note
Dieses CLIA-Kit verwendet das Sandwich-CLIA-Prinzip. Die in diesem Kit enthaltene Mikro-CLIA-Platte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes bFGF/FGF2 ist. Proben oder Standards werden in die Vertiefungen der Mikro-CLIA-Platte gegeben und mit dem spezifischen Antikörper kombiniert. Anschließend werden nacheinander ein biotinylierter Detektionsantikörper, der spezifisch für humanes bFGF/FGF2 ist, und Avidin-Meerrettichperoxidase (HRP)-Konjugat in jede Mikrotiterplattenvertiefung gegeben und inkubiert. Freie Komponenten werden abgewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Nur die Vertiefungen, die humanes bFGF/FGF2, biotinylierten Detektionsantikörper und Avidin-HRP-Konjugat enthalten, zeigen Lumineszenz. Der relative Lumineszenzwert (RLU) wird vom Chemilumineszenz-Immunassay-Analysegerät bei 425 nm gemessen. Der RLU-Wert ist positiv mit der Konzentration von humanem bFGF/FGF2 assoziiert. Die Konzentration von humanem bFGF/FGF2 in den Proben kann durch Vergleich des RLU-Werts der Proben mit der Standardkurve berechnet werden.
