Technical note
Dieses CLIA-Kit nutzt das Sandwich-CLIA-Prinzip. Die in diesem Kit enthaltene Mikro-CLIA-Platte ist mit einem Antikörper vorbeschichtet, der spezifisch für humanes Hepc ist. Proben oder Standards werden in die Vertiefungen der Mikro-CLIA-Platte gegeben und mit dem spezifischen Antikörper kombiniert. Anschließend werden nacheinander ein biotinylierter Detektionsantikörper, der spezifisch für humanes Hepc ist, und ein Avidin-Meerrettichperoxidase (HRP)-Konjugat zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Freie Komponenten werden abgewaschen. Die Substratlösung wird jeder Vertiefung hinzugefügt. Nur jene Vertiefungen, die humanes Hepc, biotinylierten Detektionsantikörper und Avidin-HRP-Konjugat enthalten, zeigen Lumineszenz. Der Wert der relativen Lichteinheit (RLU) wird mit dem Chemilumineszenz-Immunoassay-Analysator bei 425 nm gemessen. Der RLU-Wert ist positiv mit der Konzentration des humanen Hepc assoziiert. Sie können die Konzentration des humanen Hepc in den Proben berechnen, indem Sie den RLU-Wert der Proben mit der Standardkurve vergleichen.
