Technical note
Caspase 9, auch bekannt als ICE-LAP6 oder Mch6, ist eine relativ vorgeschaltete Caspase im Prozess der Apoptose-Signaltransduktion. Nachdem Mitochondrien Cytochrom c freisetzen, kann Caspase 9 einen Komplex mit Cytochrom c und Apaf1 bilden und aktiviert werden. Aktivierte Caspase 9 kann Caspase 3, das wichtigste Enzym der Apoptose, aktivieren und dadurch nachfolgende apoptotische Signale fördern. Die Aktivierung von Caspase 9 kann durch Phosphorylierung reguliert werden. Dieses Caspase-9-Aktivitäts-Assay-Kit basiert auf Caspase 9, die das Substrat Ac-LEHD-pNA katalysieren kann, um gelbes pNA (p-Nitroanilin) zu erzeugen. Das pNA hat eine starke Absorption nahe 405 nm. Die Aktivität von Caspase 9 kann durch Bestimmung der Absorption bei 405 nm berechnet werden.
