Caspase 8, auch bekannt als FLICE, MACH und Mch5, wird selbstaktiviert oder als Reaktion auf die Spaltung durch Granzym B aktiviert. Caspase 8 erkennt spezifisch IETD. Aktivierte Caspase 8 kann nicht nur PARP spalten, sondern auch an der Aktivierung anderer Proteasen der ICE-Familie, wie Caspase 3 und Caspase 7, beteiligt sein. Dieser Caspase 8 Aktivitätstestkit basiert auf Caspase 8, die das Substrat Ac-IETD-pNA katalysieren kann, um gelbes pNA (p-Nitroanilin) zu erzeugen. Das pNA hat eine starke Absorption nahe 405 nm. Die Aktivität von Caspase 8 kann durch Bestimmung der Absorption bei 405 nm berechnet werden.
