Technical note
Caspase 6, auch bekannt als Mch-2, wurde ursprünglich in menschlichen Jurkat-Zellen entdeckt. Der Vorläufer von Caspase 6 kann durch Granzym B gespalten werden, um ein aktiviertes Caspase-6-Dimer zu bilden, das Apoptose induziert. Caspase 6 kann PARP und Keratin-18 spalten, sowie Lamin A, ein wichtiges Strukturprotein der Kernhülle. Nur Caspase 6 kann Lamin A innerhalb der Caspase-Familie spalten. Dieses Caspase-6-Aktivitätstestkit basiert auf Caspase 6, die das Substrat Ac-VEID-pNA zu gelbem pNA (p-Nitroanilin) katalysieren kann. Das pNA weist eine starke Absorption bei 405 nm auf, Die Aktivität von Caspase 6 kann durch Bestimmung der Absorption bei 405 nm berechnet werden.
