Caspase 2, auch bekannt als Ich-1 (ICE und CED-3 Homolog 1) oder Nedd-2, kann während der Signaltransduktion der Apoptose aktiviert werden. Caspase 2 mRNA kann in zwei verschiedene Formen gespalten werden. Das mRNA-Produkt der Volllängen-Caspase 2 kann Apoptose fördern, während das Proteinprodukt der kurzen mRNA Apoptose hemmen kann. Caspase 2 kann in vitro durch Caspase 1, Caspase 3 und Granzym B aktiviert werden. Dieses Caspase 2 Aktivitäts-Assay-Kit basiert auf Caspase 2, das das Substrat Ac-VDQQD-pNA zu gelbem pNA (p-Nitroanilin) katalysieren kann. Das pNA hat eine starke Absorption bei 405 nm. Die Aktivität von Caspase 2 kann durch die Bestimmung der Absorption bei 405 nm berechnet werden.
