Caspase 1, auch bekannt als Interleukin 1β Converting Enzyme (ICE), ist die einzige Caspase in der Caspase-Familie, die IL-1β-Präfixe oder IL-18-Präfabrikationen spalten kann. Caspase 11 kann die 45 kD Caspase 1-Präparation spalten, um 20 kD und 10 kD Fragmente zu erzeugen, die Heterodimere bilden und durch zwei Heterodimere weiter Tetramere bilden können. Caspase 1 reguliert die Apoptose durch Spaltung von Bcl-XL und die damit verbundene Immunantwort durch Spaltung von Zytokin-Vorstufen oder reguliert die Pyroptose durch Hydrolyse von Gastermin D (GSDMD). Dieser Caspase 1-Aktivitäts-Assay-Kit basiert auf Caspase 1, das das Substrat AC-YVAD-pNA katalysieren kann, um gelbes pNA (p-Nitroanilin) zu erzeugen. Das pNA hat eine starke Absorption bei 405 nm. Die Aktivität von Caspase 1 kann durch Bestimmung der Absorption bei 405 nm berechnet werden.
