Ammoniak ist eine wichtige Stickstoffquelle für lebende Systeme und für die Synthese von Aminosäuren notwendig. Ammoniak ist ein Metabolit, der durch die Desaminierung von Aminosäuren entsteht. Es spielt eine wichtige Rolle sowohl in der normalen als auch in der abnormalen Tierphysiologie. Bei schweren Lebererkrankungen kann Ammoniak nicht aus dem Kreislauf entfernt werden, was zu einem Anstieg des Blutammoniaks führt, der eine hepatische Enzephalopathie (Leberkoma) verursachen kann. In der klinischen Praxis ist die Messung des Blutammoniaks ein wichtiger experimenteller Diagnose- und Überwachungsindex der hepatischen Enzephalopathie.
Das Nachweisprinzip dieses Kits: Die enzymkatalysierte Ammoniakreaktion verbraucht NADH, wodurch dessen Absorption bei 340 nm abnimmt. Durch Messung der Rate des Absorptionsabfalls bei 340 nm kann der Ammoniakgehalt gemessen werden.
