Citrat ist in tierischem Blut, Geweben und Pflanzenfrüchten weit verbreitet. In lebenden Organismen wird Citrat typischerweise aus Oxalacetat und Acetyl-CoA über die Citratsynthase synthetisiert. Als Zwischenmetabolit im Tricarbonsäurezyklus (TCA) kann CA unter Stressbedingungen schnell zur ATP-Erzeugung genutzt werden, wodurch die Stressresistenz des Organismus verbessert wird.
Citrat wird unter der Katalyse der Citratlyase in Oxalacetat umgewandelt. Oxalacetat wird anschließend über mehrere enzymatische Reaktionen in Wasserstoffperoxid (H2O2) umgewandelt. Das erzeugte H2O2 reagiert mit der fluoreszierenden Sonde ADHP zu einer fluoreszierenden Substanz. Die Fluoreszenzintensität kann gemessen werden, um den Citratgehalt in der Probe zu berechnen.
