Die Alpha-Glucosidase katalysiert die Freisetzung von α-Glucose vom nicht-reduzierenden Ende des Substrats und fördert die Aufnahme von Glucose durch den Dünndarm. Die Hemmung der α-Glucosidase ist eine wirksame Methode zur Behandlung von nicht-insulinabhängigem Diabetes mellitus (NIDDM).
Das Nachweisprinzip dieses Kits: Die α-Glucosidase katalysiert die Substratreaktion zur Erzeugung farbiger Produkte, deren Extinktion bei 400 nm zunimmt. Die Zugabe eines Inhibitors unterdrückt die enzymatische Aktivität der α-Glucosidase, was zu einer reduzierten Rate des Extinktionsanstiegs führt. Die Hemmrate kann basierend auf der Extinktionsdifferenz berechnet werden.
