Ferroptosis-Suppressor-Protein-1 (FSP-1) induziert eine Nicht-Caspase-abhängige Apoptose basierend auf Aminosäuren der C-terminalen Fragmente, nuklearen Translokation, Überexpression und anderen Faktoren. Der FSP-1-CoQ10-NAD(P)H-Signalweg, parallel zum klassischen Glutathion (GSH)-GPX4-Signalweg, kann Zellen vor Ferroptose schützen und spielt eine „zweischneidige“ Rolle bei den Lebensaktivitäten der Zelle.
Das Nachweisprinzip dieses Kits: FSP-1 katalysiert die Substratreaktion zur Erzeugung von Fluoreszenzprodukten, und der Inhibitor hemmt die Aktivität von FSP-1 und reduziert die Rate des Anstiegs des Fluoreszenzwertes. Die Hemmrate kann berechnet werden, indem der relative Änderungswert der Enzymaktivität bei einer Anregungswellenlänge von 535 nm und einer Emissionswellenlänge von 587 nm gemessen wird.
