Use the listed assay range, sample guidance and protocol documents to assess suitability for the intended study.

Test principle

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human APOL1. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Human APOL1. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Human APOL1, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Human APOL1 in the samples is then determined by comparing the OD of the samples to the standard curve.

Apolipoprotein L1 (APOL1; ApoL-I/APO-L) is a secreted HDL-associated protein involved in lipid-transport biology and a pore-forming component of human serum trypanosome lytic factor. It is studied in lipoprotein transport and innate trypanolytic immunity.

Biological function: Reviewed UniProt describes APOL1 as a possible participant in lipid exchange, lipid transport and reverse cholesterol transport, and as a crucial component of human trypanosome lytic factor. APOL1 forms pores in Trypanosoma brucei lysosomal membranes, contributing to parasite lysis; plasma APOL1 associates with ApoA-I-containing HDL particles.
