Les E3 ubiquitine ligases à domaine HECT transfèrent l'ubiquitine des enzymes de conjugaison d'ubiquitine E2 aux substrats protéiques, ciblant ainsi des protéines spécifiques pour la dégradation lysosomale. La protéine codée médie l'ubiquitination de multiples substrats cibles et joue un rôle essentiel dans le transport du sodium épithélial en régulant l'expression en surface cellulaire du canal sodique épithélial, ENaC. Des polymorphismes mononucléotidiques dans ce gène peuvent être associés à l'hypertension essentielle.
