La phosphatase alcaline, isozyme non spécifique tissulaire (ALPL), est une protéine membranaire appartenant à la famille des phosphatases alcalines. Il existe au moins quatre phosphatases alcalines distinctes mais apparentées chez l'homme : l'AP intestinale (IAP), l'AP placentaire (PLAP), l'AP des cellules germinales (GCAP) dont les gènes sont regroupés sur le chromosome 2, et l'isozyme non spécifique tissulaire (TNAP) dont le gène est situé sur le chromosome 1. Les phosphatases alcalines (AP) sont des enzymes dimériques qui catalysent l'hydrolyse des phosphomonoesters avec libération de phosphate inorganique. L'ALPL native est un homodimère glycosylé attaché à la membrane par une ancre GPI. Cet isozyme pourrait jouer un rôle dans la minéralisation squelettique. Des mutations du gène ALPL ont été directement liées à différentes formes d'hypophosphatasie, caractérisées par un cartilage et des os mal minéralisés, et ce trouble peut varier en fonction de la mutation spécifique, car cela détermine l'âge d'apparition et la gravité des symptômes.

ALPL (alkaline phosphatase, biomineralization associated) encodes tissue-nonspecific alkaline phosphatase (TNAP/TNSALP), a membrane-bound ectophosphatase central to extracellular phosphate metabolism and skeletal mineralization.
Biological function: The ALPL protein is the tissue-nonspecific alkaline phosphatase isozyme. It hydrolyses phosphate-containing substrates, including inorganic pyrophosphate (PPi) and pyridoxal 5'-phosphate. By reducing extracellular PPi, a mineralization inhibitor, ALPL supports hydroxyapatite formation and normal bone and tooth mineralization.
