Le kit ELISA humain AURKB (Aurora Kinase B) est un kit ELISA d'ELK Biotechnology pour la mesure d'AURKB à des fins de recherche.
Le principe de test appliqué dans ce kit est l'immunodosage enzymatique en sandwich. La plaque de microtitration fournie dans ce kit a été pré-revêtue d'un anticorps spécifique à l'AURKB humain. Des étalons ou des échantillons sont ajoutés aux puits appropriés de la plaque de microtitration, puis un anticorps conjugué à la biotine spécifique à l'AURKB humain est ajouté. Ensuite, de l'avidine conjuguée à la peroxydase de raifort (HRP) est ajoutée à chaque puits de la microplaque et incubée. Après l'ajout de la solution de substrat TMB, seuls les puits contenant de l'AURKB humain, un anticorps conjugué à la biotine et de l'avidine conjuguée à une enzyme présenteront un changement de couleur. La réaction enzyme-substrat est arrêtée par l'ajout d'une solution d'acide sulfurique et le changement de couleur est mesuré par spectrophotométrie à une longueur d'onde de 450 nm ± 10 nm. La concentration d'AURKB humain dans les échantillons est ensuite déterminée en comparant la DO des échantillons à la courbe étalon.

Biological function: AURKB is a serine/threonine kinase within the chromosomal passenger complex. It regulates chromosome alignment and segregation, kinetochore-microtubule attachment, spindle organisation and the onset and completion of cytokinesis. AURKB phosphorylates chromosomal-passenger-complex components and multiple mitotic substrates, and participates in the abscission checkpoint that prevents premature cytokinetic separation when chromosome bridges persist.
