La protéine 1 médiatrice du point de contrôle des dommages à l'ADN est une protéine de 2080 acides aminés qui, chez l'homme, est codée par le gène MDC1. La protéine codée par ce gène contient un domaine forkhead N-terminal, deux motifs C-terminaux BRCA1 (BRCT) et un domaine central avec 13 répétitions d'une séquence d'environ 41 acides aminés. La protéine codée est nécessaire pour activer les points de contrôle du cycle cellulaire en phase intra-S et en phase G2/M en réponse aux dommages à l'ADN. Cette protéine nucléaire interagit avec l'histone H2AX phosphorylée près des sites de cassures double brin de l'ADN par l'intermédiaire de ses motifs BRCT, et facilite le recrutement de la kinase ATM et du complexe protéique de recombinaison méiotique 11 vers les foyers de dommages à l'ADN.
