La mitogène-activated protein kinase kinase kinase kinase 5 est une enzyme qui, chez l'homme, est codée par le gène MAP4K5. Ce gène code un membre de la famille des sérine/thréonine protéines kinases, très similaire à la levure SPS1/STE20 kinase. La levure SPS1/STE20 fonctionne près du début des cascades de signalisation de la MAP kinase, essentielles à la réponse aux phéromones de la levure. Il a été démontré que cette kinase active la Jun kinase dans les cellules de mammifères, ce qui suggère un rôle dans la réponse au stress. Deux variants de transcription épissés alternativement et codant la même protéine ont été décrits pour ce gène.
