Le kit ELISA Human NLRP12 (protéine 12 contenant les domaines NACHT, LRR et PYD) est un kit ELISA d'ELK Biotechnology pour la mesure de recherche de NLRP12.
Le principe du test appliqué dans ce kit est l'immunodosage enzymatique en sandwich. La plaque de microtitration fournie dans ce kit a été pré-enduite d'un anticorps spécifique au NLRP12 humain. Les étalons ou les échantillons sont ajoutés aux puits appropriés de la plaque de microtitration, puis un anticorps conjugué à la biotine spécifique au NLRP12 humain est ajouté. Ensuite, l'avidine conjuguée à la peroxydase de raifort (HRP) est ajoutée à chaque puits de microplaque et incubée. Une fois la solution de substrat TMB ajoutée, seuls les puits contenant du NLRP12 humain, un anticorps conjugué à la biotine et de l'avidine conjuguée à une enzyme présenteront un changement de couleur. La réaction enzyme-substrat est arrêtée par l'ajout d'une solution d'acide sulfurique et le changement de couleur est mesuré par spectrophotométrie à une longueur d'onde de 450 nm ± 10 nm. La concentration de NLRP12 humain dans les échantillons est ensuite déterminée en comparant la DO des échantillons à la courbe étalon.
