Technical note
Ce kit ELISA utilise le principe ELISA sandwich. La plaque micro-ELISA fournie dans ce kit a été pré-enduite d'un anticorps spécifique à la GAL3 humaine. Des échantillons (ou étalons) et un anticorps lié à la peroxydase de raifort (HRP) spécifique à la GAL3 humaine sont ajoutés dans les puits de la plaque micro-ELISA. La GAL3 humaine présente dans les échantillons (ou étalons) se combine avec l'anticorps pré-enduit et l'anticorps de détection lié à la HRP spécifique à la GAL3 humaine. L'excès de conjugué et l'échantillon ou l'étalon non lié sont éliminés de la plaque par lavage. La solution de substrat est ajoutée à chaque puits. La réaction enzyme-substrat est arrêtée par l'ajout d'une solution stop, et la couleur vire au jaune. La densité optique (DO) est mesurée par spectrophotométrie à une longueur d'onde de 450 nm ± 2 nm. La valeur de la DO est proportionnelle à la concentration de la GAL3 humaine. La concentration de GAL3 humaine dans les échantillons est ensuite déterminée en comparant la DO des échantillons à la courbe étalon.
