Technical note
Ce kit ELISA utilise le principe ELISA sandwich. La plaque micro-ELISA fournie dans ce kit a été pré-enduite d'un anticorps spécifique au FE humain. Les échantillons (ou étalons) et un anticorps lié à la peroxydase de raifort (HRP) spécifique au FE humain sont ajoutés aux puits de la plaque micro-ELISA. Le FE humain contenu dans les échantillons (ou étalons) se combine à l'anticorps pré-enduit et à l'anticorps de détection lié à la HRP spécifique au FE humain. L'excès de conjugué et l'échantillon ou l'étalon non lié sont éliminés de la plaque par lavage. La solution de substrat est ajoutée à chaque puits. La réaction enzyme-substrat est arrêtée par l'ajout d'une solution stop et la couleur vire au jaune. La densité optique (DO) est mesurée par spectrophotométrie à une longueur d'onde de 450±2 nm. La valeur de la DO est proportionnelle à la concentration de FE humain. La concentration de FE humain dans les échantillons est ensuite déterminée en comparant la DO des échantillons à la courbe étalon.
