Technical note
Ce kit CLIA utilise le principe Sandwich-CLIA. La microplaque CLIA fournie dans ce kit a été pré-enduite d'un anticorps spécifique à l'OxLDL humain. Les échantillons ou les étalons sont ajoutés dans les puits de la microplaque CLIA et combinés à l'anticorps spécifique. Ensuite, un anticorps de détection biotinylé spécifique à l'OxLDL humain et un conjugué Avidine-Horseradish Peroxidase (HRP) sont ajoutés successivement à chaque puits de la microplaque et incubés. Les composants libres sont éliminés par lavage. La solution de substrat est ajoutée à chaque puits. Seuls les puits contenant l'OxLDL humain, l'anticorps de détection biotinylé et le conjugué Avidine-HRP produiront une luminescence. La valeur d'unité de lumière relative (RLU) est mesurée par l'analyseur d'immunoessais par chimioluminescence à 425 nm. La valeur RLU est positivement associée à la concentration d'OxLDL humain. Vous pouvez calculer la concentration d'OxLDL humain dans les échantillons en comparant la valeur RLU des échantillons à la courbe étalon.
