Technical note
Ce kit CLIA utilise le principe Sandwich-CLIA. La microplaque CLIA fournie dans ce kit a été pré-enduite d'un anticorps spécifique à l'hepcidine humaine (Human Hepc). Des échantillons ou des étalons sont ajoutés aux puits de la microplaque CLIA et combinés à l'anticorps spécifique. Ensuite, un anticorps de détection biotinylé spécifique à l'hepcidine humaine et un conjugué avidine-peroxydase de raifort (HRP) sont ajoutés successivement à chaque puits de microplaque et incubés. Les composants non liés sont éliminés par lavage. La solution de substrat est ajoutée à chaque puits. Seuls les puits contenant de l'hepcidine humaine, l'anticorps de détection biotinylé et le conjugué avidine-HRP apparaîtront luminescents. La valeur de l'unité de lumière relative (RLU) est mesurée par l'analyseur d'immunodosage par chimioluminescence à 425 nm. La valeur RLU est positivement associée à la concentration d'hepcidine humaine. La concentration d'hepcidine humaine dans les échantillons peut être calculée en comparant la valeur RLU des échantillons à la courbe étalon.
