Dieses Gen gehört zur Homeobox-Genfamilie. Die Homeobox-Gene kodieren eine hochkonservierte Familie von Transkriptionsfaktoren, die eine wichtige Rolle bei der Morphogenese in allen vielzelligen Organismen spielen. Säugetiere besitzen vier ähnliche Homeobox-Gencluster, HOXA, HOXB, HOXC und HOXD, die sich auf verschiedenen Chromosomen befinden und aus 9 bis 11 tandemartig angeordneten Genen bestehen. Dieses Gen ist eines von mehreren Homeobox-HOXD-Genen, die sich in einem Cluster auf Chromosom 2 befinden. Deletionen, die den gesamten HOXD-Gencluster oder das 5'-Ende dieses Clusters entfernen, wurden mit schweren Extremitäten- und Genitalanomalien in Verbindung gebracht.
