Rekombinantes Maus-Komplementkomponente C3a wird durch unser E. coli Expressionssystem hergestellt und das Zielgen, das Ser671-Arg748 kodiert, wird exprimiert.
Das Komplementsystem wird als Schlüsselkomponente der angeborenen Immunität und als erste Verteidigungslinie im Kampf gegen eindringende Krankheitserreger definiert. Komplement 3 (C3) ist die am häufigsten vorkommende Komponente der Komplementkaskade und der Konvergenzpunkt für alle drei Hauptaktivierungswege des Komplementsystems: nämlich den klassischen, den alternativen und den Mannose-bindenden Lektinweg. Die Komplementaktivierung führt zur Bildung der C3-Konvertase, die C3 in die wichtigen Effektormoleküle C3a (Anaphylatoxin) und C3b (Opsonin) spaltet, die dann die Entfernung von Mikroben vorantreiben. Durch die Bindung an den C3a-Rezeptor (C3aR) zeigt C3a eine potente Anaphylatoxin-Aktivität, einschließlich erhöhter Gefäßpermeabilität, Auslösung der Degranulation von Mastzellen, Entzündung und Aktivierung von Leukozyten.
