Das Coronavirus-N-Protein wird für die RNA-Synthese des Coronavirus benötigt und besitzt RNA-Chaperon-Aktivität, die am Template-Switch beteiligt sein könnte. Das Nukleokapsidprotein ist ein am häufigsten vorkommendes Protein des Coronavirus. Das N-Protein verpackt die positiv-strängige virale Genom-RNA in ein helikales Ribonukleokapsid (RNP) und spielt eine grundlegende Rolle bei der Virion-Montage durch seine Interaktionen mit dem viralen Genom und dem Membranprotein M. Es spielt eine wichtige Rolle bei der Verbesserung der Effizienz der subgenomischen viralen RNA-Transkription sowie der viralen Replikation. Aufgrund der Konservierung der N-Proteinsequenz und ihrer starken Immunogenität wird das N-Protein des Coronavirus als Diagnosetool ausgewählt.
