Rekombinante humane Rezeptor-Tyrosin-Protein-Kinase ErbB-2 wird von unserem Säugetier-Expressionssystem produziert und das Zielgen, das Thr23-Thr652 kodiert, wird mit einem 6His-Tag am C-Terminus exprimiert.
Der humane epidermale Wachstumsfaktorrezeptor 2 (HER2) ist eine Art von Membranglykoprotein und gehört zur Familie der epidermalen Wachstumsfaktor (EGF)-Rezeptoren. HER2 spielt eine Schlüsselrolle bei der Entwicklung, Zellproliferation und -differenzierung. Es wurde berichtet, dass HER2 mit Malignität und einer schlechten Prognose bei zahlreichen Karzinomen assoziiert ist, einschließlich Brust-, Prostata-, Eierstock-, Lungenkrebs und so weiter. HER2 wird durch Dimerisierung aktiviert und nicht durch EGF, TGF-alpha und Amphiregulin. Die Interaktion mit PTK6 erhöht seine intrinsische Kinaseaktivität. Es bildet Heterodimere mit EGFR, ERBB3 und ERBB4. HER2 assoziiert mit der 5'-TCAAATTC-3'-Sequenz im PTGS2/COX-2-Promotor und aktiviert dessen Transkription. Es ist an der transkriptionellen Aktivierung von CDKN1A beteiligt und die Funktion des Proteins beinhaltet STAT3 und SRC. Und es ist auch an der Transkription von rRNA-Genen durch RNA Pol I beteiligt und fördert die Proteinsynthese und das Zellwachstum.
