Rekombinanter Maus Insulin-ähnlicher Wachstumsfaktor I wird von unserem E. coli-Expressionssystem produziert und das Zielgen, das Gly49-Ala118 kodiert, wird mit einem 6His-Tag am C-Terminus exprimiert.
Der Insulin-ähnliche Wachstumsfaktor I (IGF1) gehört zur Familie der Insulin-ähnlichen Wachstumsfaktoren, die strukturell homolog zu Proinsulin sind. Maus-IGF-I wird als zwei Vorläufer-Isoformen mit alternativen N- und C-terminalen Propeptiden synthetisiert. Diese Isoformen werden von verschiedenen Geweben unterschiedlich exprimiert. Reifes Maus-IGF-I teilt 94% und 99% Aminosäuresequenzidentität mit menschlichem bzw. Ratten-IGF-I und weist eine speziesübergreifende Aktivität auf. Es teilt 60% Aminosäuresequenzidentität mit reifem Maus-IGF-II. IGF-I induziert die Proliferation, Migration und Differenzierung einer Vielzahl von Zelltypen während der Entwicklung und postnatal. Es spielt eine wichtige Rolle bei der Muskelregeneration und dem Tumorwachstum. IGF-I bindet an IGF-I R, IGF-II R und den Insulinrezeptor. Die Assoziation von IGF-I mit IGF-Bindungsproteinen erhöht seine Plasmahalbwertszeit und moduliert seine Wechselwirkungen mit Rezeptoren.
