Das Ratten-CRP(C-reaktives Protein) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von CRP.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Ratten-CRP ist. Standards oder Proben werden dann mit einem biotin-konjugierten Antikörper, der spezifisch für Ratten-CRP ist, in die entsprechenden Mikrotiterplattenvertiefungen gegeben. Als Nächstes wird mit Meerrettichperoxidase (HRP) konjugiertes Avidin zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen, die Ratten-CRP, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-CRP in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
