Das Maus-CTLA4(Cytotoxic T-Lymphocyte Associated Antigen 4) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsbestimmung von CTLA4.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus-CTLA4 ist. Standards oder Proben werden zu den entsprechenden Vertiefungen der Mikrotiterplatte hinzugefügt, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für Maus-CTLA4 ist. Anschließend wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Maus-CTLA4, biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-CTLA4 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.

Biological function: CTLA4 (CD152) is an inhibitory immune receptor expressed by activated T cells and regulatory T cells. It binds the B7 ligands CD80 and CD86 and counterbalances costimulatory signalling, limiting T-cell activation.
