Das Zebrafisch 5-HT(5-Hydroxytryptamin) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von 5-HT.
Dieser Test verwendet die kompetitive Enzymimmunoassay-Technik. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit Zebrafisch 5-HT vorbeschichtet. Standards oder Proben werden den entsprechenden Vertiefungen der Mikrotiterplatte hinzugefügt, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für Zebrafisch 5-HT ist. Anschließend wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Danach wird TMB-Substratlösung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Zebrafisch 5-HT in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
