Der Maus MIP4a (Makrophagen-Inflammationsprotein 4 Alpha) Mikrosampel ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Erforschung und Messung von MIP4a.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Antikörper vorbeschichtet, der spezifisch für Maus MIP4a ist. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, danach wird ein biotin-konjugierter Antikörper, der spezifisch für Maus MIP4a ist, hinzugefügt. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Maus MIP4a, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus MIP4a in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
