Der Humane SPX(Spexin) ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Messung von SPX in der Forschung.
Das in diesem Kit angewandte Testprinzip ist ein Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem für humanes SPX spezifischen Antikörper vorbeschichtet. Standards oder Proben werden zu den entsprechenden Vertiefungen der Mikrotiterplatte hinzugefügt, anschließend ein biotin-konjugierter Antikörper, der spezifisch für humanes SPX ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen eine Farbveränderung, die humanes SPX, biotin-konjugierten Antikörper und enzym-konjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem SPX in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
