Das Rinder-FGF2 (Fibroblast Growth Factor 2, Basic) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von FGF2.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Rinder-FGF2 ist. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen gegeben, dann mit einem biotin-konjugierten Antikörper, der spezifisch für Rinder-FGF2 ist. Als Nächstes wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die Rinder-FGF2, den biotin-konjugierten Antikörper und das enzymkonjugierte Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Rinder-FGF2 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
