Das Ratten-NMU(Neuromedin U)-ELISA-Kit ist ein ELK Biotechnology ELISA-Kit zur Messung von NMU in der Forschung.
Dieser Assay verwendet die kompetitive Enzymimmunoassay-Technik. Die in diesem Kit enthaltene Mikrotiterplatte ist mit Ratten-NMU vorbeschichtet. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, anschließend wird ein Biotin-konjugierter Antikörper hinzugefügt, der spezifisch für Ratten-NMU ist. Danach wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Anschließend wird TMB-Substratlösung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-NMU in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
