Das Ratten-APOA5(Apolipoprotein A5) Mikrosample ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Forschungsbestimmung von APOA5.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Ratten-APOA5 ist. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für Ratten-APOA5 ist. Als Nächstes wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die Ratten-APOA5, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-APOA5 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.

APOA5 (apolipoprotein A5) encodes apolipoprotein A-V (ApoA-V), a liver-enriched minor apolipoprotein associated mainly with HDL and also with triglyceride-rich lipoproteins. APOA5 is an important regulator of plasma triglyceride metabolism and is studied in lipoprotein lipase activity, VLDL handling and systemic lipid homeostasis.
Biological function: Apolipoprotein A-V is a minor plasma apolipoprotein that modulates triglyceride-rich lipoprotein metabolism. Reviewed UniProt describes ApoA-V as a potent stimulator of apo-CII-dependent lipoprotein lipase triglyceride hydrolysis and as an inhibitor of hepatic VLDL-triglyceride production, supporting efficient triglyceride clearance and regulation of circulating triglyceride levels.
