Das Ratten-PCSK9 (Proprotein Convertase Subtilisin/Kexin Typ 9) Microsample ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von PCSK9.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper beschichtet, der spezifisch für Ratten-PCSK9 ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, anschließend wird ein biotin-konjugierter Antikörper, der spezifisch für Ratten-PCSK9 ist, hinzugefügt. Als Nächstes wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die Ratten-PCSK9, den biotin-konjugierten Antikörper und das enzymkonjugierte Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-PCSK9 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
