Das Maus-COL6(Kollagen Typ VI)-ELISA-Kit ist ein ELK Biotechnology ELISA-Kit zur Forschungsmessung von COL6.
Das in diesem Kit angewendete Testprinzip ist ein Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus-COL6 ist. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für Maus-COL6 ist. Anschließend wird Avidin, das mit Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Maus-COL6, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Schwefelsäurelösung beendet und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-COL6 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
