Das Maus NUCB1 (Nucleobindin 1) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von NUCB1.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus NUCB1 ist. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für Maus NUCB1 ist. Als Nächstes wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die Maus NUCB1, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus NUCB1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
