Das Human PCMT1 (Protein L-Isoaspartate-O-Methyltransferase) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von PCMT1.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes PCMT1 ist. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, anschließend ein biotin-konjugierter Antikörper, der spezifisch für humanes PCMT1 ist. Danach wird zu jeder Mikrotiterplattenvertiefung Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbveränderung, die humanes PCMT1, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem PCMT1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
