Das Pig ICAM1 (Intercellular Adhesion Molecule 1) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology für die Forschungsmessung von ICAM1.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Schweine-ICAM1 ist. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen gegeben, dann mit einem biotin-konjugierten Antikörper, der spezifisch für Schweine-ICAM1 ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die Schweine-ICAM1, einen biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Schweine-ICAM1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
