Der Maus S100A7(S100 Calcium Binding Protein A7) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsbestimmung von S100A7.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem spezifischen Antikörper gegen Maus S100A7 vorbeschichtet. Standards oder Proben werden dann zusammen mit einem Biotin-konjugierten Antikörper, der spezifisch für Maus S100A7 ist, in die entsprechenden Mikrotiterplattenvertiefungen gegeben. Als Nächstes wird Avidin, das mit Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen, die Maus S100A7, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten, einen Farbwechsel. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und der Farbwechsel wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus S100A7 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
