Das Ratten-FABP3 (Fatty Acid Binding Protein 3, Muskel und Herz) Mikrosample ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Erforschung und Messung von FABP3.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Ratten-FABP3 ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, dann mit einem biotin-konjugierten Antikörper, der spezifisch für Ratten-FABP3 ist. Anschließend wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Ratten-FABP3, einen biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-FABP3 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
