Das Rat AST (Aspartat-Aminotransferase) Microsample ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von AST.
Das in diesem Kit angewandte Testprinzip ist ein Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Ratten-AST ist. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für Ratten-AST ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Ratten-AST, einen Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-AST in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
