Der Humane TACTILE(T-Cell Activation Antigen, Increased Late Expression) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von TACTILE.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Humanes TACTILE ist. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, anschließend mit einem Biotin-konjugierten Antikörper, der spezifisch für Humanes TACTILE ist. Als Nächstes wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die Humanes TACTILE, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Humanem TACTILE in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
